I am installing a new 750gig HD with Vista Home Premium in a new computer(DIY). What is the consenus these days on partitioning, is it wise, and will Vista install process accomplish it?

Logical Drives
Of course, you need to create at least a single partition in order to use your drive.
I believe the Vista installer still allows you to create a boot partition of the size you desire, and then partition the rest of the drive later.
I myself like to use multiple logical drives.
That way, I can store my data files separate from my OS and can easily back up just my files.
