You can try this
First from server manager disconnect all active users.
Then search for all files with LCK extention (*.lck) and (to be on the safe side just rename thew extentions)delete the ones associated with the database file.
reboot and then try opening the database.
(Be sure to be logged on as administrator while doing so)

I had the exact same problem today with the server service, here's what you do.

1. Check the event log for failed services.
2. Go to services under control panel and set all non-critical services to manual start, and stop the services, this is of course after notifying users that the server will be offline. Make notes of the orginal service startup configurations.
3. Remove any offending services. I use Srvinstw.exe from NT 4.0 Res kit Supplement 4.
4. Reboot.
5. Reinstall any servicesremoved in step 3. Do not reboot yet.
6. Reapply the CURRENT service pack.
7. Reboot.
8. Restore all services to their original start states as per the notes from step 2
9. Reboot to unsure all services restart correctly.
10. Recheck the event viewer for success.
