Check some of your firewall, malware or similar programs that you have installed to see if they block modification of the HOSTS file, which is common practice.

Also allot of malicious ware will edit your hosts file to redirect you to their BIG SALE ON VIRUS REMOVAL.

I would try opening your run command and your cmd prompt if you cannot you probably got some form of infection.

    next step boot in safemode
    run msconfig
turn off anything that looks suspect while running a Malwarebytes scan as well as what ever Antivirus you have

Also for the record spybot locks out the hosts file because it puts known offensive sites on your localhost so that they are never resolved through dns.

This is also a rather effective way to block out Facebook and PronoTubedotnet and other sites you might find unsavory, but if you have allot to blacklist this can get extensive.

There is also Open DNS service which can be either free or has plans you can buy that will block known offensive sites from being resolved as well also you can log what gets surfed and searched from your IP.

If you are logged on as a normal user (which you should be under normal circumstances) you won't have access.
