Reverse Mappings on a Proxy Server

How does the Reverse Mappings work on a Proxy Server? Also in general how does a proxy server works? We are trying to surface a web based application through a shared (by more than one application) proxy server but are having URL mapping problems (acording to our network staff). Application server’s response fails to reach the user’s browser.
