Transaction Log in MS-SQL Data base

We are using SAP ECC 5.0 and using MS-SQL 2000 as our backend. Our Transaction Log file has touched 85 GB whereas our DB size is approx. 65 GB. We are not able to perform many activities and not able to take backup of Transaction Log also. What should be done? Please help
