DEFRAG always in SAFE MODE?

i understand executing defrag in safe mode if i have winme or any other windows where swapfile is in same partition as windows.
if swapfile is on a second hdd, do i have to preceed the same way(defrag in safe mode) or can i do this in normal windows?
