3-tier web-application design?

I need a broad view on how to devlop a 3 tier web based app using Microsoft technologies. I know the tech aspects of VB and ASP and COM and HTML and all and have developed app for myself. now I need to deisgn a app for a client and so i needthe best pratices/ guidelines.. some framework… some typical implementations…
