NT Defrag

I have an NT 4.0 SP6a machine box with one harddrive. The drive is partitioned. Drive c: is formatted fat 16 and holds the OS. Drive D: is formatted NTFS. Can boot from a DOS disk and run the DOS defragger on the C: drive?
