Delete a single roaming profile???

I want to save roaming profiles on local machine…However I do periodically want to delete some profiles and not others/all. How come some will delete and others will not? I get an error of: Cannot delete Desktop.ini: Access is denied. The source file may be in use.

Any suggestions will be greatly appreciated.
