Exchange 2000: SMTP stop all outgoing emails!

Running SBS2000 with Exchange 2000, we use POP for incoming emails and SMTP using an external relay to send out. Last Thursday without any apparent reason all outgoing emails stopped working. All emails are queued in the SMTP Outgoing queue which promptly goes into “retry”. The relay host (Gradwell) have tested our connection and said all is fine, I have sent them message Headers, but they say they only show internal activity, ie the message doesn’t get as far as the Relay server.
I have TELNET to local address and sent EHLO, this only returns (of the X- codes) x-link2state and x EXCH50. According to MS this would point to IIS not working correctly.
I have run ISINTEG -FIX (which fixed a few errors), but still no joy, run ESEUTIL /G, but no negative results.
I am now backing up Exchange in order to reinstall it: can anyone help me before I get into rebuilding Exchange? Please help!
