Intermittent Permissions

I have a new SBS 2003 server with latest service packs. Its experiencing intermittent permissions on its directories. One day a user can access the file and the next they cannot. The primary directory grants everyone change access and then inside the directory are individual folders with security settings only granting specific users access. Any idea what could cause this?
