Permission problems on IIS

I am running an IIs server hosting 5 websites.

The sites are all runing fine execpt for one.
Suddenly the server extentions have no write permission to a file. I checked C:\inetpub\www.mysite.com and I can’t change the permissions on all the files for that website.As an administrator It comes out access denied to a lot of the files.
