removal of virus..

how do i remove virus without formating my systems hardisk because of virus my system has become very slow and when i start the system then it takes too much time may be half an hour to show the desktop..?:(
although i have scaned it with semantic…also tell me which anti virus is best and from which URL i can download the trial version of antivirus.
