Scheduling Backup

In Windows NT Server i am having 5 Disk partions. I want to have BackUp Scheduling for all the 5 Disk’s at 23:00 Hrs. At the time of Backup, is it necessary to stop all the services. And in Windows NT Server i am having Ms Exchange Server as front end for user applications.
