Using partitions efficiently

What is the best way to arrange applications, system files & OS, data and my files across the multiple partitions? I have seen a few small DOS-based apps for partitioning drives but they seem to be designed for quite high-level users and admins. Simply, what is the best way to partition my drive and make the best use of the facility?
