Have all files contained within folders on a external drive been opened?

A directory of all folders and files contained on a personal external drive has been created. The directory originally had an md5 file extension. It was later changed to a txt file extension. The directory document was then zipped so that it could be delivered via e-mail. In this case, the directory document is 5,283 pages long. If a directory document like this was created, does it then mean that every file on the personal external drive that contains all of the individual files, has been opened and reviewed? If I only have a txt file document of the directory for the personal external drive, is there any way to tell whether each file was opened as a part of creating the directory of the external drive?
