2000 Server Permissions

I am fairly new at managing permissions as you will probably be able to tell by my question, but here goes. When you are setting permissions on shared folders, where is the best place to set those permissions? You have permissions that you can set under the sharing properties and also permissions under Security. Do you normally give everyone rights under sharing permissions and then manage access rights through the Security tab or vice versa? Thanks in advance
