Damn good question! You should ALWAYS have a backup of AD in the event you have a disaster.

For Windows 2000, you can use the steps documented here to backup Active Directory:

http://www.microsoft.com/technet/prodtechnol/windows2000serv/technologies/activedirectory/maintain/opsguide/part1/adogd03.mspx

For Windows 2003, I recommend the free ebook by NetIQ, which has a number of administrative activities documented:

http://www.windowsitlibrary.com/ebooks/administeringad/Index.cfm

Microsoft's Windows 2003 documentation on backup can be found here:
http://www.microsoft.com/resources/documentation/WindowsServ/2003/standard/proddocs/en-us/Default.asp?url=/resources/documentation/WindowsServ/2003/standard/proddocs/en-us/ntbackup_restore_sysstate.asp

Basically, you want to make a backup of the system state and the SYSVOL as documented here:

http://www.microsoft.com/resources/documentation/WindowsServ/2003/standard/proddocs/en-us/Default.asp?url=/resources/documentation/WindowsServ/2003/standard/proddocs/en-us/ntbackup_backup_sysstate.asp
