Basically it means that the same tape isn't picked up after it has been written to.

The Bar codes on the spines of the tapes allow a series to be deployed without any mix up.

I have yet to get my hands on an autoloader. how does it pick the tape when you ask for one? if you put the wrong tape in that spot, will it reject it before it reads the tape?

The barcode simply allows the tape management software to keep track of the individual cartridges. The management software allows the user to specify which volume(s) to use for a particular backup purpose. The barcodes also enable the tape library to track the physical location of each cartridge, allowing automated mounting / dismounting of the cartridges, when called for by the backup software.
