Big defragmenation problem with 2000Server domain controller

We have a 2000Server Domaincontroller. boot and systempartion are on one drive. A second hard drive contains Powerquest Multicast software and a lot of images (in .pqi format)
After noticing that restoring images has slowed down I inspected the state of fragmentation on this harddrive (230 GB) and found 50 – 60% fragementation. So i decided to Deragment with the intenal Windows tool from ‘Disk manager’ The tool has finished but i LOST my shers to the drive!. after rebooting Diskmanager says Disk ‘healthy’ but it’s no lnger formatted so i cannot acces it. It looks “all my data has lost”
Recover programs cannot restore the data.

The issue is that i did exactly the same defragmentation on a other separate system that contains the same configuration (Domain controller and seperate 230GB data drive)That acts as spare system.
Unfortunally the result was the same ALL DATA on the second harddrive lost after deragmentation. I have solved very lot of problems. But such as this never happened
I have a big disaster ! Can anyone help me????
