CHANGE FROM FAT TO NTFS

Can I copy a fat partition for NT C: drive to another drive that is NTFS? And can I then switch the drive letters around and create a new C: drive and label the old C: drive as H or I drive or whatever the next drive letter is. Doing this would I now be able to boot up with the new C: drive being NTFS??
