How can I restrict certain PC's within my intranet to one specific internet URL?

In IE under tools\internet options\content, you can enable the Content Advisory and restrict the sites there.

Yes. You can use Content Advisory in IE. You can use IEAK (Internet Explorer Administration Kit to create policies which can be installed in IE. You even can create "Automatic Configuration Script" and point required IE to it (Tools->Internet Options->Connections->LAN Settings). After that you can change settings in one centralized place and these settings will be propagated to computers when IE starts next time. But in case you have "advanced" users - only solution is mandatory proxy/firewall with filters.
