I have the following on my computer (Windows XP), taking up much space:
Microsoft .NET Framework 1.1 Microsoft .NET Framework 2.0 Service Pack 2 Microsoft .NET Framework 3.0 Service Pack 2 Microsoft .NET Framework 3.5 SPI
Which of these -- if any -- do I really need? What happens if I uninstall any or all of them? I do very little development work now, and none (as far as I know) using .NET Framework in any way. Could these be used as run-time resources for other applications?

It's basically a giant runtime environment, like Java or the much smaller Visual Basic or Visual C runtimes.
Applications should tell you in their system requirements whether they need .NET. The ultimate way to find out is to go ahead and uninstall the whole ball of wax, and if some application you like or need whines about it, you can install just the version required. (Download from Microsoft.) I don't know if Windows Update would attempt to automatically reinstall it or not, but if it does, there should be a way to refuse the .NET.
Disclaimer: I've never had any .NET on on any XP machine.
I have the same concerns where I'm trying to speed up my computer. Which files are you saying to backup? The Net.Framework files or what? I tried to uninstall an old version when the latest version showed up after automatic updates. However I couldn't delete the old version.
Deleting this isn't really going to speed up anything, unless your are out of disk space. Make sure you don't need it for anything. You probably need to be an Administrator to uninstall.
If the uninstaller is broken, use something other than Windows add/remove. (CCleaner, Revo Uninstaller, whatever.)
