What can I do to determine the cause of my server slowdown

I have recently replaced an older 2003 server with a Dell 2950 with dual gigabyte nic’s 4 gb of ram and 2 quad processors. The server is fine until more than 20 users login and start to work. There are only two applications on this server. One is a Microsoft Access database that only a few use and the other is a database app that most of the company uses. I suspect the server is too fast for the rest of the network. All the the switches are 10/100 switches. All the pc’s connecting are Windows XP and some users come in through a Windows 2000 terminal server. Utilization goes from 2% up to 65% and no one can do their job. I can’t see anything in the event logs that would indicate the problem. Can someone help, please?
